Fast, accurate, reliable AAV quantification using Amperia, a comparison with ELISA

The extensive manual handling required for AAV quantification methods such as ELISA, not only delays the time to results but also increases the risk of errors and variability.

This study compared the accuracy, reproducibility and efficiency of the Amperia Redox Electrochemical Detection (RED) system with traditional ELISA techniques when measuring AAV capsid titre. Tests were repeated in duplicate, across a range of concentrations, over three days by different operators.

The study demonstrates Amperia RED as a robust alternative to ELISA for fast, accurate convenient AAV quantification.

Key takeaways

Accurate and reliable

  • Amperia demonstrated greater accuracy and consistency in measurements at each sample concentration and across three test days.
  • Amperia RED results fell within a 6% average error range from the known concentration, compared to up to a16% for the ELISA test results.
  • The Amperia system can quantify a broad range of AAV concentrations with an automated process increasing consistency.
  • The ELISA protocol requires serial dilution and manual handling throughout, increasing the risk of error and variation between operators.

Reduced hands-on time, faster time to result

  • Amperia requires just 10 minutes hands-on time per run (compared with 4-5 hrs. for ELISA).
  • Actionable analysed results are available around 3 x faster than ELISA.
  • Amperia supports quicker decisions and enhanced workflow efficiency.
